In recent years, Targeted Protein Degradation (TPD), as a novel way to induce protein degradation, has become a new drug discovery strategy. This technology utilizes a naturally occurring degradation mechanism (UPS or ALP) in cells to regulate the degradation of disease-causing target proteins, which is expected to break through the bottleneck of traditional drug development and provide a new chemical tool and drug discovery strategy for the field of molecular targeted therapy. This new type of drug development also shows great promise in oncology, neurodegenerative diseases, autoimmune diseases, inflammatory diseases, and viral and bacterial infections. Up to now, dozens of targeted protein degraders have entered the clinical stage, and the number of clinical trials is growing at a rapid rate every year. Despite the rapid development of targeted protein degradation technologies, there are also some technical difficulties and challenges that need to be addressed, such as toxicity, drug resistance and limited degradation range.
